Definitely not.  It is not the referee's role to "enforce" playing time 
policy.
See below.

For reference: AYSO National policy requires that all players play half
of the time they are present at the match.

The "3 before 4" playing time guideline (no player plays 4 quarters before
all players have played 3 quarters) is a strong recommendation, but not
currently a strict policy, in region 45.  It is a regional policy in 
some regions
in our area.  (It may become region 45 policy in the future.)

It is appropriate for a referee to remind a coach - for example, "coach, 
are you aware
player #12 has already sat out one period?"  That should be the extent 
of the referee's
or assistant referees' involvement with this issue, during the match.

There may be totally fair and valid reasons why certain players play 
less than three
periods on a particular weekend. The referee team at any one match 
cannot know
all the background for the coach's decisions (nor should it).  The 
referee team's role
is to observe and report.

Make sure that player participation time is correctly recorded on the 
lineup cards,
and entered correctly into WebYouthSoccer.

Make a note in the "game comments" section of WebYouthSoccer if you 
think it is
called for.

The division coordinators and the region board monitor the WebYouthSoccer
data for patterns of the same players playing half the game while other 
players
play the entire game, week after week, and will intervene when appropriate.
